Isotretinoin works by influencing cell cycle progression, differentiation, survival, and apoptosis, which collectively reduce sebum production. This action helps prevent clogged pores and limits the proliferation of acne-causing bacteria. Both isotretinoin and its metabolite, 4-oxo-isotretinoin, significantly decrease sebum production. Isotretinoin exhibits minimal binding to retinol-binding proteins (RBPs) and retinoic acid nuclear receptors (RARs), while tretinoin and 4-oxo-tretinoin selectively bind to the RAR-γ receptor, which may contribute to its anti-acne properties. By inducing apoptosis in sebocytes, it further reduces sebum levels. It also minimizes comedone formation by reducing hyperkeratinization through an unknown mechanism. Although isotretinoin does not directly kill bacteria, it creates an environment less favorable for bacterial growth by shrinking sebum ducts. Additionally, it may enhance immune responses and alter monocyte chemotaxis to reduce inflammation.

- Overdose has not been reported with Retigel. However, accidental ingestion could lead to symptoms of hypervitaminosis A, including severe headaches, nausea, vomiting, drowsiness, irritability, and itching.
✔️ Side Effects of Retigel 0.05% Topical Gel
- Common side effects include stinging, burning, or irritation at the application site.
- Erythema and peeling may occur.
- If excessive irritation develops, temporarily stop using the gel and resume once symptoms subside. If irritation persists, discontinue use altogether.
