Tacrolimus, a macrolide immunomodulator derived from Streptomyces tsukubaensis, functions by inhibiting T-cell activation. It binds to the intracellular protein FKBP-12, forming a complex that blocks calcineurin activity, thereby suppressing the transcription of pro-inflammatory cytokines and downregulating the immune response.
