✔️ Mode of Action

Progesterone, primarily secreted by the corpus luteum, induces secretory changes in the endometrium, promotes mammary gland development, relaxes the uterus, blocks follicular maturation and ovulation, and helps maintain pregnancy.

✔️ Dosage: Prinagel Vaginal Gel

Oral Progestogen (for Menopausal Hormonal Replacement Therapy)

• Adult: 200 mg daily as a single dose at night for 12-14 days per month.

Amenorrhea

• Adult: 400 mg daily for 10 days.

Dysfunctional Uterine Bleeding

• Adult: 400 mg daily for 10 days.

Prevention of Endometrial Hyperplasia

• Adult: 200 mg taken orally at night for 12 days in a sequential 28-day cycle.

Vaginal Gel for Dysfunctional Uterine Bleeding

• Adult: 45 mg every other day from the 15th to the 25th day of the cycle. If no response, increase the dose to 90 mg.

Vaginal Gel for Amenorrhea

• Adult: 45 mg every other day from the 15th to the 25th day of the cycle. Increase the dose to 90 mg if necessary.

✔️ Administration of Prinagel Vaginal Gel

Each applicator contains 90 mg of progesterone.

1. Open the tube with the sharp point and attach the applicator.
2. Squeeze the tube to fill the applicator up to the ring mark.
3. Insert the applicator into the vagina while lying down.
4. Push the plunger completely to release the gel.
5. Clean the applicator with warm soapy water after use.

No known interactions with other drugs, but Prinagel should not be used with other vaginal treatments.

✔️ Overdose Effects of Prinagel Vaginal Gel

Acute overdose is unlikely, but in case of overdose, discontinue use and treat symptomatically.

✔️ Pregnancy & Lactation

• Pregnancy: Safe during pregnancy (Category A). Used successfully for embryo implantation and pregnancy maintenance.
• Lactation: Not recommended during breastfeeding, as progesterone may be present in milk.

• Family history of breast cancer, deep vein thrombosis (DVT), or pulmonary embolism (PE).
• Current or history of depression, endometriosis, diabetes, hypertension, renal/hepatic impairment, or bone metabolic diseases.
• Conditions worsened by fluid retention (e.g., migraine, asthma, epilepsy).
• Discontinue if jaundice, visual problems (including contact lens intolerance), or signs of thromboembolic events occur.
• Discontinue 4 weeks before major surgery or prolonged immobility.
• Use with caution in patients on warfarin or oral anticoagulants (may require dose adjustment).
• Long-term use may increase the risk of breast, cervical, and liver cancers.
• Use cautiously in postmenopausal women over 65 due to possible links to dementia.
• Increased risk of developing breast cancer and cervical cancer, with long-term oral contraceptive use.
